dAb Screening Platform

In Alphamab, we have established phage library derived from naive or immunized camel. Through HTS biopanning, the target specific domain antibodies (dAb) can be quickly enriched and further characterized.

Through collaboration with College of Life Science, Southeastern University, Alphamab has optimized a technology platform to use phage display to screen dAb library from immunized camel. The platform covers antigen prep, camel immunization, PMBC library construction and HTS. 

1. Soluble antigen preparation. Based on optimized protein engineering and production systems (E.Coli and 293 cell), the antigen with native folding and bioactivity can be produced in short time.

2. Optimized library assembly. Routinely we are getting the primary immunized library at 10E7 to 10E8 and higher than 99% diversity. The high quality library guaranteed the good hit with sub nM to pM affinity, by pass the labor and time consuming affinity maturation step in most case. 

3. Efficient and reliable dAb screening and characterization. We have developed multiple screening schemes, including direct screening, negative screening, competition and cell based screening. Those schemes can be tailored to different antigens and target antibody profile. Each leads obtained will be further selected through a tiered evaluation system to assess its drug like property and manufacturability.

So far, we have already successfully obtained dAbs against several targets. 

In addition, Alphamab has collaborated with Xinjiang University to make most diversified naive dAb library at 10E9. The library will be applied to certain high value targets that are not amenable for immunization. With help from top notch computational and structural biologists, we are going to further develop the naive library into focused library, biased for agonist or antagonist, for GPCRs, ion Channels and other membrane protein targets.
